  HYPOGLYCEMIC PROPERTIES OF LEAF EXTRACTS DERIVED FROM MISTLETOES IN ALLOXAN-INDUCED DIABETIC RATS

Abstract:

Folk medicine traditions suggest the use of certain mistletoes for managing diabetes in Nupeland. In this study, we investigated the hypoglycemic effects of aqueous and ethyl acetate extracts from two mistletoe species, Globimetula braunii and Globimetula oreophila, in diabetic rats. Diabetes was induced intraperitoneally in rats using 100mg/kg body weight of alloxan, and rats with blood glucose levels exceeding 135 mg/kg body weight were considered diabetic. The study comprised twenty-one rats, divided into seven groups of three rats each. Diabetic rats were administered 500mg/kg body weight of the respective extracts once daily for fourteen days. Blood glucose levels were monitored every five days and showed a significant reduction during the treatment period compared to the untreated diabetic group (DNT). The experiment lasted for twenty-one days, after which blood serum and fresh organs were collected following euthanasia.

The extracts demonstrated significant (P<0.05) hypoglycemic effects, with reductions of 58.37%, 50.46%, 65.50%, and 50.65% for Aqueous Globimetula braunii (AGB), Aqueous Globimetula oreophila (AGO), Ethylacetate Globimetula braunii (EGB), and Ethylacetate Globimetula oreophila (EGO), respectively. There were no significant (P>0.05) changes in body weight in any of the groups treated with the extracts, except for the normoglycemic group (NNT), which exhibited a significant (P<0.05) increase of 8.04% in total body weight. The mean percentage ratio of fresh organs to whole body weight for the liver and heart did not show significant (P>0.05) differences between the groups. However, the lungs and pancreas exhibited significant (P<0.05) reductions in all groups treated with the extracts. The kidney of the AGB-treated group showed a significant (P<0.05) increase compared to the NNT group. There were no significant differences (P<0.05) in serum total protein and cholesterol levels between the groups. Only the Diabetes untreated group (DNT) showed a significant (P<0.05) increase (173.50+16.57) in triglyceride levels compared to the NNT group. Among the groups, only the EGO group showed a significant (P<0.05) increase (1.57+0.2) in the level of direct bilirubin, while total bilirubin levels did not significantly (P>0.05) differ from the NNT group. The AGB group showed a significant increase in the activities of alkaline phosphatase. EGB, EGO, and DNT showed significant (P<0.05) increases (78.90+8.90, 63.30+6.10, and 73.07+2.40, respectively) in the activities of alanine aminotransferase compared to the NNT group. All groups showed significant (P<0.05) increases in the activities of aspartate aminotransferase compared to the NNT groups. In conclusion, our research demonstrates that the extracts of G. braunii and G. oreophila at a dosage of 500mg/kg body weight exhibit significant anti-diabetic effects in a diabetic animal model (rats), while also ameliorating the biochemical abnormalities associated with diabetes.
